ABSTRACT

The aim was to evaluate the impact of supplementation with different protein sources from agro-industrial production on the parasitological profile of ovine matrices and the development of their offspring. The protein sources used were: soybean bran, cotton pie, babassu pie, and coconut pie. A total of 60 ovine matrices were used, with confirmed pregnancy and supplemented in the final third of gestation at 0.5% of live weight in pastures of deferred Marandu grass. The evaluations occurred from the final third of gestation and in the lactation phase until weaning. The data collected in the matrices were: counting of eggs per gram of feces (EPG) and identification of infective larvae (Faecal test), evaluation of the degree of anemia by the FAMACHA© method, body weight (kg), and body condition score (BCS). In the lambs, the biometric measurements were taken from the 30 days of life until weaning. In Faecal test, the prevalence was 98% of Haemonchus spp. There was no effect of the protein sources for EPG (3476.0) and FAMACHA©. However, in the mats supplemented with coconut pie were observed the lowest body weights (36.9 kg) and lower BCS (2.7). Babassu, coconut and cotton cakes, and soybean meal used as a protein source in the supplementation of ovine matrices kept in deferred pastures of Marandu grass at the 0.5% level of live weight maintain the production indexes consistent with the physiology stage of the matrices even with a high endo-parasitic load.

ABSTRACT

Abstract The aim here is to present data on the efficacy of anthelmintics in sheep flocks in Rio de Janeiro, Brazil, and to discuss the interpretation of the fecal egg count reduction test (FECRT) for each nematode genus. Fecal eggs counts and pre- and post-treatment coprocultures were performed, the former to evaluate the efficacy of and the latter to determine the overall parasite prevalence. An additional efficacy test was performed at Farm # 1 a year after the initial test. Severe anthelmintic resistance was found for the flocks, with no FECRT sensitivity at any of the 22 farms evaluated. However, an analysis of the infective larvae showed that some drugs were effective against certain parasitic genera; i.e., levamisole was more effective against Haemonchus spp. and moxidectin against Trichostrongylus spp. In the additional FECRT performed at Farm # 1, moxidectin and nitroxynil were ineffective separately, but when applied in combination they were highly effective due to their efficacy against Haemonchus (nitroxynil) and Trichostrongylus (moxidectin), respectively. The use of the FECRT targeting the parasitic nematode species prevalent on farms may make it possible to choose more effective anthelmintics.

ABSTRACT

Abstract The Neotropical green-belly stink bug, Dichelops melacanthus (Dallas) is a major pest of maize, Zea mays L. in the main production areas of Brazil. It usually feeds on the stems of young plants (seedlings) causing heavy damage by affecting the plant growth and mitigating seed yield. Laboratory studies were conducted to determine body position (upward or downward) of the bug on plant (seedling) stem during feeding and not feeding activities. Ten visual records were taken per day, each spaced one hour during 30 days of 10 adult bugs of similar age exposed to maize seedlings inside cages (plastic tubes). At each observation, it was recorded if the bug was feeding on the stem (i.e., stylets inserted into the plant tissue) or not, and its body position. During feeding, waveforms were recorded using the EPG (electropenetrography) technique, which were correlated with histological studies to reveal the feeding sites. Results indicated that when they were feeding, the majority of the bugs were in the downward position. In contrast, when the bugs were on the plants, and not feeding, they were mostly in the upward position. Waveforms generated using the EPG coupled with histological studies demonstrated that during ingestion bugs fed from the xylem vessels and from the parenchyma tissue using cell rupture strategy in the latter. No clear explanation was found to explain the preferred downward body position during ingestion, but some hypothesis are speculated.

ABSTRACT

PURPOSE: To develop a novel combination of controlled aliasing in parallel imaging results in higher acceleration (CAIPIRINHA) with integrated SSFP (CAIPI-iSSFP) for accelerated SSFP imaging without banding artifacts at 3T. MATERIALS AND METHODS: CAIPI-iSSFP was developed by adding a dephasing gradient to the balanced SSFP (bSSFP) pulse sequence with a gradient area that results in 2π dephasing across a single pixel. Extended phase graph (EPG) simulations were performed to show the signal behaviors of iSSFP, bSSFP, and RF-spoiled gradient echo (SPGR) sequences. In vivo experiments were performed for brain and abdominal imaging at 3T with simultaneous multi-slice (SMS) acceleration factors of 2, 3 and 4 with CAIPI-iSSFP and CAIPI-bSSFP. The image quality was evaluated by measuring the relative contrast-to-noise ratio (CNR) and by qualitatively assessing banding artifact removal in the brain. RESULTS: Banding artifacts were removed using CAIPI-iSSFP compared to CAIPI-bSSFP up to an SMS factor of 4 and 3 on brain and liver imaging, respectively. The relative CNRs between gray and white matter were on average 18% lower in CAIPI-iSSFP compared to that of CAIPI-bSSFP. CONCLUSION: This study demonstrated that CAIPI-iSSFP provides up to a factor of four acceleration, while minimizing the banding artifacts with up to a 20% decrease in the relative CNR.

ABSTRACT

Enumeration of nematode eggs in fecal samples using the McMaster technique and morphological identification of third stage larvae from fecal cultures have been extensively used with satisfactory results in the diagnosis of the gastrointestinal nematode infections in ruminants. In order to improve sensitivity and accuracy, other approaches for quantification of eggs have been employed, like the FLOTAC and Mini-FLOTAC techniques. Results obtained in different studies indicate that fecal egg counts are a reliable measure of the size of the worm burden. However, the immunological status of the animals should be taken into consideration to interpret the results of the fecal examination. Molecular techniques have also been useful in the diagnosis of parasitic diseases. The ultimate in diagnosis has been the development of robotic platforms that enable separation of eggs from feces. Because manipulation is minimal, good quality DNA from eggs is obtained, which is used for amplification, and finally, produces a result indicating the degree of the infection by the different parasite species in mix infections. The ideal method should be reliable, friendly to non-experts and quick to perform. With the advance in robotics, bioinformatics and molecular biology, methods with such characteristics are expected to become available and affordable to be used in laboratories for the routine diagnosis of gastrointestinal nematodes of ruminants...

ABSTRACT

In this study, we aimed to establish the prevalence and risk factors relating to gastrointestinal helminthiasis, and to characterize the sanitary management practiced among sheep herds in the Sertão region of the state of Paraíba, northeastern Brazil, based on factors that condition the ways of controlling these parasites in these herds. The research was carried out between April and July 2012. We visited 54 farms, where fecal and blood samples were individually collected from 465 animals. On each farm, a questionnaire was applied to gather information on variables relating to potential risk factors. The prevalence of sheep gastrointestinal helminthiasis in the region was 75.9%. At least one animal tested positive for this helminthiasis on 53 (98.1%) of the 54 farms evaluated. The eggs per gram of feces (EPG) analysis showed the following infection burdens: 51.8% with mild infection, 27.1% moderate infection, 9.9% heavy infection and 11.2% fatal infection. Among the sheep farms visited, anthelmintics were used on 81.5% (p <0.05). The most relevant risk factor in this study was the farm area, because it defines the area available for grazing animals. Properties with many animals and little pasture area, which are the most abundant type in the Sertão region of Paraíba, tend to have high prevalence of gastrointestinal helminthiasis, because the animals are more prone to reinfection. The Sertão region of Paraíba presents high prevalence of gastrointestinal helminthiasis among sheep, and the farm area is the most relevant risk factor for the development of these parasites.

ABSTRACT

The aim of this work was to evaluate the effect of silicon and the insecticide dimetoate in the probing behavior of the greenbug Schizaphis graminum (Rond.) (Hemiptera: Aphididae) in wheat plants. The treatments were: 1) silicon in soil, 2) silicon in the soil plus foliar spraying, 3) silicon foliar, 4) silicon soil plus insecticide, 5) silicon in soil plus foliar and insecticide, 6) foliar spraying of silicon and insecticide, 7) insecticide and 8) untreated. The data was compared by the multifactorial analysis of variance for each one of the factors separately. The percentage data was analyzed by the test of Chi-Square. It was not verified interaction of silicon and dimetoate and, also, of the isolated effect of silicon in the probing behavior of the greenbug. However, in plants treated with the dimetoate, a larger percentage of insects presented the xylem phase and lower number of aphids reached the phloem. In general, the application of dimetoate changed the probing behavior of the S. graminum.

ABSTRACT

The aim of this cross-sectional observational study was to determine the frequency and factors associated with infection by Eimeria spp. and gastrointestinal nematodes in 356 calves on 20 dairy farms located in southern Minas Gerais, Brazil. Ten species of Eimeria spp. were identified, of which E. bovis (37.6%) and E. zuernii (17.9%) were the most frequent. From fecal cultures, four genera of gastrointestinal nematodes were recovered, of which Cooperia spp. (74.6%) and Haemonchus (19.4%) were the most frequent. Variables relating to higher levels of technology used on dairy farms showed a significant association (p < 0.05) with higher OPG and EPG counts, and are discussed in this study.

ABSTRACT

Greenidea ficicola Takahashi and Greenidea psidii van der Goot (Aphididae: Greenideinae) are Asian aphid species newly introduced in Brazil associated with Moraceae and Myrtaceae. The feeding behavior of G. ficicola and G. psidii was investigated on their respective host plants, Ficus benjamina (Moraceae) and Psidium guajava (Myrtaceae), using the Electrical Penetration Graph (EPG). Fifteen females of each aphid species were monitored during 24h using a DC-EPG GIGA-4 monitor. The time spent in phloem phase (waveforms E1 and E2) was 13.6 percent of the total recording time for G. ficicola and 0.8 percent for G. psidii. The average time in the pathway phase (waveforms C and pd) represented 50 percent of the total time for both species. Aphids spent more time in non-penetration and stylet pathway activities than in the phloem phase or actual feeding. The principal component analysis (PCA) showed that the two species formed different groups in relation to EPG parameters, despite some overlapping. The probing patterns with multiple penetrations of short duration in the sieve elements for both species may indicate apparent unsuitability for sustained feeding on their respective host plants. These results suggest that these two exotic species are in the process of adaptation to their host plants in their new environment and/or the plants may present either chemical or physical barriers against these insects.

ABSTRACT

Chemotherapy of clonorchiasis with praziquantel (PZQ) is effective but about 15% of treated cases have been reported uncured. The present study investigated correlation of single nucleotide polymorphisms (SNPs) of the cytochrome P450 gene, CYP3A5 and cure of clonorchiasis. A total of 346 egg passing residents were subjected and treated by 3 doses of 25 mg/kg PZQ. Reexamination recognized 33 (9.5%) uncured and 313 cured. Numbers of eggs per gram of feces (EPGs) before treatment were significantly lower in the cured group than in the uncured group (2,011.2+/-3,600.0 vs 4,998.5+/-7,012.0, PA and g.27526C>T of CYP3A5 were 15.2% and 9.1% while those were 3.8% and 1.0%, respectively, in the cured group. The cure rate was significantly lower in the cases with SNP at g.27526C>T and EPGs> or =1,000. In conclusion, EPGs and SNPs of CYP3A5 are factors which influence cure of clonorchiasis by PZQ therapy. It is strongly suggested to recommend 2-day medication for individuals with high EPGs> or =1,000.

ABSTRACT

Aphids are the most important vectors of viruses infecting potato (Solanum tuberosum). We focused on the response of the aphid vector Myzus persicae (Sulzer) to five commercial potatocultivars: Ágata, Jaette Bintje, Mondial, Monalisa and Santè, by traditional antibiosis and antixenosis tests and by the EPG (Electrical Penetration Graph) technique, as a step forward to the design of effective management practices. Our aim was to identify plant factors involved in resistance of these cultivars against M. persicae, both at the surface and in deeper plant tissues. Results from the antixenosis test confirmed a strong preference of M. persicae for the Mondial cultivar. The antibiosis study indicated a lower population development of the aphid in 'Monalisa' when compared to 'Ágata' and 'Jaette Bintje'. EPG assays indicated that 'Santè' inhibited the initial feeding process of M. persicae, whereas 'Monalisa' showed a physical-type of resistance as demonstrated by a very high number of short probes. The cultivar Mondial showed average values for all EPG variables analyzed. The behavior in 'Jaette Bintje' indicated this cultivar was an ideal host for aphid feeding and reproduction. Together, the EPG data revealed the existence of pre and post-phloematics factors in the cultivars under study, which have important implications on the efficiency of transmission and spread of virus in potato by M. persicae.

ABSTRACT

The control of helminth infections has an essential role in the productivity of dairy cattle. Sustainability, cost/benefit and feasibility of the chosen control measures, must be taking into account for each production system. This study aimed to establish criteria for anthelmintic treatment of dairy cattle, in different categories, based on eggs per gram of feces (EPG) counting. For this purpose, it were analyzed fecal samples from 100 dairy cattle, in different age groups, from small farms of the Central region of Rio Grande do Sul state, Brazil. Based on these results, the selective treatments, based on EPG, are indicated to female calves (until three months old) and lactating cows, and the strategic control to male and female calves, from four to 24 months old.

ABSTRACT

Quantitative fecal egg counts represented as the number of eggs per gram of feces (EPG) are generally a reliable parameter to estimate the worm burden of intestinal and hepatic parasitoses. Although Haplorchis taichui (Digenea: Heterophyidae) is one of the most common minute human intestinal flukes, little is known about the relationship between EPG and the actual worm burden in patients or the severity of the disease. In the present study, fecal samples were collected from 25 villagers in northern Thailand before and after praziquantel treatment. The EPG values of each participant were determined by the modified cellophane thick smear method, and adult worms were collected from the whole stool after the treatment. Eggs per day per worm (EPDPW) of H. taichui were estimated 82 from egg counts and expelled worms. The EPG was not well correlated with the worm burden, and a reverse correlation was observed between the EPDPW and the worm burden.

ABSTRACT

The Kato-Katz (KK) method is a well-known method of fecal examination for helminthiases. Its diagnostic sensitivity was found very high for clonorchiasis. The present study evaluated the correlation of Clonorchis sinensis egg counts by the KK method with those by direct smear and formalin-ether (FE) technique. The egg counts obtained by the KK method (Y) were correlated with the counts by direct smear (X) with the equation of Y = 659.4 + 0.266X (r2= 0.738), but not with those by the FE method. The present study demonstrated that the KK method and direct smear were useful for both qualitative and quantitative diagnosis of clonorchiasis, especially in the field.
